Question: Describe about the Fixation and Tissue Processing for Anatomic Dissection. Answer: Introduction The preparation of tissues for experimental purpose involves the anatomic dissection of rats. The tissues taken from the rat are of trachea, oesophagus, lung, duodenum, liver, kidney and femur. The tissues obtained from the dissection are separated from the rat and are subjected to irrigation by normal saline after the division into smaller pieces for the experiment. Then the fixation of the tissues is done by keeping the tissues in 10% Normal Buffered Formalin (NBF), (1) which also helps in preserving the cells and tissues and maintains the viability of the cells and tissues. The tissue processor machine fixes the tissues and then performs the processing. The steps involved in processing are as following; the tissues are dehydrated by increasing the alcohol concentration. Care should be taken tom prevent the tissues from damage. Therefore the concentration of alcohol is raised slowly and effectively. To remove the alcohol from the tissues and prevent the alcohol from reaching in the tissues, Xylene agent is used. The Femur tissues mainly consist of calcium salt and to prepare the tissues for the processing it is important to decalcify the tissue. As the completion stage is attained for the tissues, it needs to be embedded in right proportion of the cassette to perform the sectioning. The tissues in the histological stage need to be placed properly in the cassette. The sectioning of the tissues is performed after the solidification of the blocks. The aim of sectioning is to obtain slice of tissues having a thickness similar to one cell. The microtome used for the sectioning should be placed at 10m, and the blocks should be snipped for obtaining the clear view of the given specimen. Then the microtome thickness should be adjusted to 4m, as it is the standard measurement for daily diagnosis. After the block preparation the tissues are put in the water bath having a temperature about 37 degrees Celsius. This helps removing the rigidity of the folds in the specimen before placing them on the microscopic slide. Haematoxylin (2) is used to stain the acidic nucleus in the specimen. Eosin is used to stain the muscles and collagen having basic properties. The HE method is preferred to get the differentiated tissue sections and to facilitate the morphological study of the tissues. Special stains are required for the investigation and for the diagnosis of liver, duodenum, oesophagus, trachea kidney. Some of these attribute are the weight, mass, volume, area (surface area), density, length, etc. This attributes can be measured but there are many units of measure involve in each of the attributes. Standard units of measure are developed so that the confusion between what unit of measure to be used is resolved (Jones, 1998). The following are the standard unit of measure for a certain attribute: Weight – Kilograms Mass – Kilograms Volume – Liters Area – square meters Length – meters As said above, measurement is a way of obtaining data. One cannot obtain the right data without the right tool to be used. As with measurement, a right measurement instrument must be used in order to get the right data and in order to estimate with precision and accuracy the object that you want to measure. The following attributes that are stated can be measured with the use of the right measuring instrument. For clarification, weight is not the same as mass. Weight is the amount of mass that is present when it is suspended in a spring scale when acted upon by the earth’s gravity. On the other hand, mass is the amount of matter present in an object. Weight is measured with the use of spring scale or a weighing scale. The value of the weight in the earth can be the same as the value of the mass. But once the force acting on the object changes the value of the weight will change and the mass will stay the same. Generally, masses like mass of a person can be obtained by the use of the same instrument used in measuring weight in earth. Volume can be measured by the use of some known instruments like measuring cup for liquid, pipette for liquid, graduated cylinder (for liquid and solid) and pneumatic trough for gases. Area is measured by the use of a planimeter. Length is usually measured by the use of instruments like ruler, tape measure, meter stick, yard stick etc (Fuchs, 1996). The attributes being measured is deeply related to the unit of measure being used. First, the attributes maybe too large or too small for unit of measure, in that case there are other units of measure given. The hierarchies of units are given in a unit system. There are two unit systems that are popular today. The SI system and the English system give a complete list of all the unit of measures according to the quantity (Abbot, 2003). Some of the attributes of the bucket can only be obtained by the use of measuring devices. Other attributes, however, can be obtained with the use of some formulas. For example: Volume and area can be obtained by formulas like V = 1/2? r2h, and A = 2? rh + ? r2. The formula for the volume is obtained by the use of the formula for the volume of a cylinder. Bucket is usually cylindrical in shape. The formula for the area is obtained by using the formula of the surface area of an open cylinder. Other formulas can be obtained for the attributes of the bucket. Another example is the density of the bucket. In general, the density is equal to the mass divided by the volume of the bucket. Since mass can be obtained with the use of a measuring device and volume can be obtained with the use of the given formula then one can solve for the density of the bucket with the general formula (Manura, 2005). Another concept that should be discussed is approximation and estimation. As said earlier, measurements are always an estimate. Approximation and estimation are very important concepts regarding measurements because they will be the basis of the accuracy and precision of a measurement. One does not get accurate and precise results all the time with the use of measuring devices. There might be errors that can come up from time to time. To properly record results obtained using measuring devices, one must take note of the errors in measurements and put it in the resulting measure to obtain a more precise and accurate result. Results should be written in the form: result + uncertainty. For example, if a buckets height according to a 9 different rulers is 20 centimeters. Uncertainty and volatility are common attributes of today's national economies. While Small and Medium Enterprises (SMEs) represent major players in these economies, their stability and profitability are crucial issues that need further investigation. This statement is particularly significant in the Saudi Arabian context, because such enterprises have relatively recently begun to emerge and are beginning to play a dominant role in the country's economy. At the same time, the SME sector in Saudi Arabia is currently facing many barriers related to their relatively new economic situation (Kuada, 2006), as the atmosphere of the economic world is constantly changing and tends to be somewhat uncertain. As a result, companies, including SMEs, need to be more creative and flexible as they seek to get maximum profits in the minimum amount of time. Moreover, it has been noted that human resources play a vital role for every company to achieve its goals (Barney & Wright, 1998), hence most companies seek to recruit high aptitude managers who can be a valuable addition to a workplace and can lead a company to achieve sustained success. In this respect, employee retention is considered one of the most serious challenges facing the stability of small organizations, since employee turnover can be harmful and expensive. When an employee intends to leave the company voluntarily, the issue is of such importance that numerous researchers have investigated the causes and effects of voluntary turnover (Al-A'Raj, 1989). Whilst the cost of losing skilled employees can be harmful for any organization (Ballinger, Craig, Cross, ; Gray, 2011), it is particularly so in SMEs as the effect is proportionally greater. According to O'Connell and Mei-Chuan (2007), the average cost of employee turnover is approximately $13,996 per employee. Further, Ballinger, Craig, Cross and Gray (2011) estimate that the direct cost of hiring and training a new employee can range from 25% to 500% of the employee's annual salary, and, as such, Boushey and Glynn (2012) found that the replacement of highly educated executive employees averages 213% of the annual salary bill for a company. It has been noted that because this significant turnover is such a costly process (Al-A'Raj, 1989), it could lead to serious negative consequences for an organization. Indeed, in some situations, the consequences of significant turnover rates could expand beyond the organization to influence the labour market, the economic cycle and communities in general. Furthermore, Hyson (2016) and George (2015) argue that an organization can both conserve knowledge and manage resources more cost-effectively through being proactive in attempting to retain valuable employees. As a result of such deliberate actions to maintain staff, organizations are likely to increase their overall performance and develop long-term stability. In this respect, it has been suggested that manager behaviour plays a vital role in staff turnover. For example, Snyder and Lopez (2009) have specifically asserted that the encouragement of skilled employees by managers is vital in this regard. Of particular interest to the current study is that, in the SME sector, because enterprises are fiscally smaller their resources are generally scarcer. In the light of this scarcity, employee turnover is a more harmful occurrence than in large organizations which are able to absorb the consequential additional financial outlay described above. For this reason, the SME sector constitutes an important area for further research. Moreover, researchers analysing the behaviour of managers in SMEs report that the manager's emotional intelligence is of great importance, as it significantly affects the management processes and outcomes within their enterprises. It is telling that the debate over topics related to EI and employee's behaviour is ongoing in different cultures and parts of the world (Ang et al., 2007) , and it is asserted here that the role of EI in reducing turnover is one particular area that requires further investigation. Therefore, it is postulated that the analysis of the role of EI in the SME context should be carefully investigated in order to understand the way it affects employee's retention outcomes in Saudi Arabia. It has also emerged that there is an extensive amount of controversy surrounding whether there are any differences in the levels and perceptions of EI in managers from different backgrounds and cultures, and, in addition, to what extent EI can be affected by these factors. limited research has been conducted on the relationship and/or link between EI and employee turnover in SMEs, specifically in the Middle Eastern context. Thus, this research aims to fill an important void in the literature.

The Green Revolution moved Indonesia from being a major rice importer in the 1970s to being self-sufficient by the mid 1980s. Based on the population quickly increasing, Indonesia is finding ways to make food production more efficient and cheaper to support the population. Indonesia had the most rapid decrease in poverty due to the fast agricultural growth created by the Green Revolution. The production of crops became quicker and cheaper, allowing the sales price to decrease. Poverty stricken people now do not have to devote their whole paycheck towards paying for food, since the sales price has lowered. Contract Law Case Study of Hotel Contract law involving a hotel and client A. In this scenario, the first significant point is the nature of the parties’ respective first dealings with one another. From the hotel’s point of view, their first contact with A was through their advertisement on their website. This advertised the price of accommodation at the Scarborough Palms Hotel as being  £300. From A’s point of view, his first contact with the hotel is through an initial emailed enquiry. Although we are not told exactly what A’s initial enquiry was concerned with, it is probable that it was simply asking for details of the offer. In order to establish what the nature of the ultimate contract is, between the hotel and A, it is first necessary to find the ‘offer’ and ‘acceptance’; the constituent parts of any contract. An offer has been held to be a statement which objectively (I.e. to a reasonable observer) indicates that the person making the ‘offer’ is prepared to contract on the terms specified in that offer (Gibson v Manchester City Council). It would at first sight, appear that the hotel’s website and advert for the accommodation at the specified price was an offer. This, however, is not the case, as it has been held by the courts that advertisements are usually ‘invitations to treat’ rather than offers, as the advert usually lacks the other essential ingredient of a contract; an intention to be legally bound (Partridge v Crittenden). This principle is in place in order to protect the advertiser from incurring liability in contract to everyone who is willing to purchase the goods (in this case, the holiday), at the advertised price. An ‘invitation to treat’ is an invitation to the other party to negotiate the terms of a potential contract. A responds to this invitation by making his initial email enquiry, which can similarly be classed as an invitation to treat, or perhaps simply an enquiry. No offer has yet been made by either party (Fisher v Bell). The hotel then respond to A’s initial enquiry informing A of a special promotion that will entitle him to the accommodation at the price of  £200. This communication will certainly be counted as an offer, as it displays an intention to be bound by the terms it mentions (a requirement that was first set out in the seminal case, Carlill v Carbolic Smoke Ball Co). A then ‘accepts’ the offer by filling in the online booking form. The hotel have stipulated a means of acceptance, by providing the online booking form which A is required to complete. This is, then, the hotel’s prescribed method of acceptance. A, through no fault of his own, is unable to complete this prescribed method of acceptance, despite his attempt. It is uncertain whether this will affect his ‘acceptance’. In Manchester Diocesan Council for Education v Commercial and General Investments Ltd, it was held that the prescribed method of acceptance was not the only possible one, prov ided the other method was no less advantageous to the offeror. Unless the hotel specifically stated that the online booking form was the only method of acceptance, A’s posting of a hard copy would be valid. We come to the issue of communication of the acceptance to the offeror. Upon A’s arrival at the hotel, he is informed that his booking form did not arrive, and that there are no available rooms. It is an established principle that an acceptance must be communicated to the offeror in order for their to be a contract (Holwell Securities Ltd v Hughes). In the present instance, however, the acceptance has been posted by A. Following Household Fire Insurance v Grant, the acceptance is effectively communicated on posting (the so called ‘postal rule‘). In order for this rule to apply, however, it must have been reasonable for A to use the post to accept the offer (Quenerduaine v Cole). In this instance, since the online booking form was not working, it seems likely that it would be found to have been a reasonable method of acceptance. As of this point, then, the contract exists between A and the hotel. The terms of the contract specify that A will have accommodation at the hotel for the price of  £200. He will later be able to claim damages for breach of contract from the hotel when they cannot furnish him with a room at that rate. Before that arises, however, there is a further contract which requires clarification. Upon A’s arrival, and discovery that his booking form has not arrived, he threatens to sue the hotel. The manager’s response is to make an offer of accommodation for  £250. It is possible that subsequently, the hotel could claim the offer was made under undue influence. It is an established principle of contract law that where a contract is induced by undue pressure, it is voidable (Williams v Bayley). This means that if the hotel can establish that the manager made the offer under pressure, the agreement could be cancelled. A agrees to these terms, and a second contract exists betwe en the parties. A then spend his holiday at the hotel and completes the second contract. A subsequently claims for damages in the amount of  £50. The hotel counter-claims for damages because A has breached the contractual term not to pursue an action against the hotel. These two claims, however, refer to two distinct contracts, and both, it would seem, are legitimate claims. It is possible that through forming the second contract, the equitable principle of waiver came into play. That is, by making the second contract, the parties (and A in particular), waived the right to claim damages (Hughes v Metropolitan Railway). There is also the possibility that by forming the second contract, the initial contract was frustrated, as it became impossible to carry out because the parties had subsequently contracted to the same agreement on different terms (Nickoll Knight v Ashton Edridge Co). With regard to the second contract, the hotel are within their rights to claim damages for A’s breach. He has clearly broken his contractual promise not to pursue an action against the hotel. There is a contract rule that a claimant cannot recover damages in respect of a loss which is too remote a consequence of the defendant’s breach of contract. If the losses flow naturally from the breach, which in this case they would appear to, the losses are recoverable (Hadley v Baxendale). The hotel, then, will be able to recover damages for A’s breach of the contract term subject, of course, to proving that this agreement was a term incorporated into the second contract. In Leslie Marmon Silko’s Ceremony, the use of storytelling is quite prevalent. Within the framework of Ceremony there are references of the tradition of Native American storytelling along with the progression of telling a story. Storytelling within the Native American culture is oral, traditionally. The method of storytelling within Ceremony at the beginning lays down the framework of the entire book. Silko starts out the novel with a series of stories. The first of which is about Ts’its’tsi’nako (or more easily said) â€Å"Thought Women†, who thinks of things and they appear. She happens to be thinking of a story and it just so happens to be the story being told to us. This then leads us to the next story (1). The next story turns out to be a story about stories. This story tells us (the reader) the importance of stories, and that they aren’t merely for entertainment, but are used to fight off death and illness. The narrator then states, â€Å"You don’t have anything, if you don’t have the stories.† Thus telling us the true importance of the stories of Native American culture, seeing as everything was passed down orally, and not much was written down if any at all (2). Now, for Tayo, these stories embody the understanding of the Native American world Tayo grew up with. Only the army, the doctors, and the white schools try to convince Tayo that the stories are wrong. As Tayo recreates and recalls the old tales, he begins to reunite with the community, pulls through the trauma of war, and ultimately brings back the rain to his land. Tayo learns from these stories that he is not alone, because the stories are shared within a community, and because the contents of the stories show him that others have shared like experiences (Notes/Class Discussions). The rest of the stories within the text of Ceremony announce elements that will reoccur within the novel. As the story is told either by a single person or by a group of people, it can fashion between those people a sense of community. As stated previously, that stories have the power to fight of death and illness. The stories contain the ceremonies and rituals that have the ability to cure individuals and the communities. Stories are able to provide this ability by restoring the affiliations betwixt all things and people. The stories within Ceremony are an integral part of the story line. The stories within the novel show us how important they are to the Native American culture and way of life. They provide us with the impending points of the plot, of how a ceremony is what can cure the people.
